FISH signal from transgenic tobaccoFISH signal from transgenic tobacco (Nicotiana tabacum) containing four copies of the pine drought-inducible lp5 gene. Using karyotypic features to help identify chromosome homologues, one locus (1) is homozygous, and two are heterozygous (2 and 3). The nontransgenic homologue of 2 is karyotypically identifiable [(2)], whereas that of 3 is not unequivocally so. The results illustrate feasibility of routine detection of alien transgenes inserted into eukaryotic genomes. Signal was detected with sequential applications of mouse anti-biotin and Cy3-conjugated goat anti-mouse antibodies. Chromosomes were counterstained with DAPI.
